Are you currently facing challenges such as high viral mutation rates, difficulty in achieving high-titer neutralization in assays, or complex structural stabilization requirements for vaccine development? Our Rotavirus VP7 Specific Neutra™ Antibody Products help you develop highly specific therapeutic candidates and streamline clinical evaluation through innovative B-cell cloning and high-precision epitope-targeting technologies that stabilize the critical VP7 trimer.
Rotavirus, a member of the Reoviridae family, remains a leading cause of severe, dehydrating diarrhea in infants and young children worldwide, contributing to approximately 200,000 deaths annually. The virus is a non-enveloped, triple-layered particle. The outermost layer is primarily composed of VP7 (Viral Protein 7), a 37-kDa calcium-binding glycoprotein, and the spike protein VP4.
Structure and Function
VP7 exists as a homotrimer on the virion surface, stabilized by two calcium ions at each subunit interface. Its primary biological function is to lock the VP4 spikes into place and maintain the structural integrity of the triple-layered particle (TLP). Once the virus enters the host cell endosome, the low calcium environment triggers the dissociation of the VP7 trimer. This "uncoating" event is a mandatory step for the release of the transcriptionally active double-layered particle (DLP) into the cytoplasm.
Signaling and Pathogenesis
VP7 interacts with host cell receptors, including integrins (e.g., α2β1 and αvβ3) and Hsc70, triggering PI3K/Akt and MAPK/ERK pathways to facilitate membrane penetration. Crucially, VP7-mediated calcium depletion from the ER activates sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) and induces autophagy via the CAMKKβ-AMPK pathway. This localized Ca2+ signaling also modulates NF-κB and interferon responses, creating a cellular environment conducive to viral replication and assembly.
The clinical impact is significant; rotavirus destroys mature enterocytes at the tips of the villi in the small intestine, leading to malabsorption and osmotic diarrhea. Furthermore, the non-structural protein NSP4 acts as a viral enterotoxin, further disrupting intracellular signaling and ion transport.

Clinical Relevance
VP7 defines the G-serotype (glycoprotein) of the virus. To date, at least 27 G-types have been identified, with G1, G2, G3, G4, and G9 being the most prevalent in human infections. Neutralizing antibodies targeting the epitopes on VP7 are a major correlate of protection against rotavirus disease, making it a primary focus for both live-attenuated and subunit vaccine strategies.
Anti-VP7 neutralizing antibodies serve as critical components in both diagnostic and therapeutic research frameworks:
