Description | This product is a monoclonal antibody derived from mouse, which can specifically recognize 5-methyl-2'-deoxycytidine (m5dCyd). The mab reacted strongly with m5dCyd and weakly with 5-methylcytidine (m5Cyd) but not with cytidine (Cyd) and 1-methyladenosine. The IC50 of the antibody IgG against m5dCyd was 4.27 ug/mL and that of scFv was 0.054 ug/ml, determined by inhibition ELISA. |

Application Notes | Measurement of DNA methylation levels To establish the inhibition ELISA-based measurement system for the DNA methylation level in cancer cell lines and healthy peripheral blood cells, the detection limit and inter- and intra-assay variation of the assay system were determined. From the standard curve using authentic m5dCyd, the detection limit was 3 ng/ml. Good linearity was seen from 7 to 300 ng/ml. The inter- and intra-assay coefficients of variation were 5.611.7% and 1.95.5%, respectively. These results show that this ELISA system was suitable for the determination of m5dCyd within the range of 7-300 ng/ml. |
