Application Notes | /b>Immunohistochemistry assay Immunohistochemistry was performed on slides of 4% buffered formalin fixed paraffin embedded samples. After deparaffinization, all slides were subjected to antigen retrieval by boiling in 10 mM citric acid supplemented with 0.05% Tween-20 (pH 6.0) at 120°C for 10 min, subsequently quenched, blocked and incubated overnight at 4°C with 0.2 to 0.5 μg/mL diagnostic monoclonal mouse anti-CLDN18.2 antibody clone 43-14A. Antibody binding was visualized with horseradish-peroxidase-labeled secondary antibodies and a substrate-chromogen solution. Sections were subsequently counter- stained with Mayer's haematoxylin and subjected to evaluation by the raters. |
