Amyloid beta (Aβ) is derived from non-amyloidogenic proteolysis of the amyloid precursor protein by β- / γ-secretase. Aβ has various forms such as Aβ monomer, Aβ oligomer, and Aβ amyloid plaques (see Figure 1). The deposition of Aβ is a critical event in the development of Alzheimer's disease (AD).

Biological Functions of Aβ

The different forms of Aβ interact with different cell surface receptors via the extracellular domain and activate downstream pathways (figure 2). These pathways play pivotal roles in the progression of Alzheimer's disease.

Aβ-based Clinical Strategies

Inhibition of Aβ oligomerization is the main traditional strategy for the treatment of AD by overexpressing β-degrading enzymes, using small molecule inhibitors of β- / γ-secretase, or blocking amyloid channels. Currently, Aβ antibodies that hydrolyze specific aggregates or reduce amyloid loads have developed into novel immunotherapy approaches for AD.

Table 1 provides some biological antibodies targeting Aβ and their ideal immunotherapy effects such as plaque clearance and reduced neurotoxicity.
