BK Virus VP1 Specific Neutra™ Antibody Products

Product list

Are you currently facing challenges such as BK virus-associated nephropathy (BKVAN) in transplant recipients or the lack of standardized high-potency tools for neutralizing assays? Our BK virus VP1 Specific Neutra™ Antibody Products help you accurately quantify viral neutralization and develop next-generation therapeutics through our proprietary Neutra™ platform and high-affinity epitope targeting of the major capsid protein.

Introduction of the BK virus VP1

BK virus (BKV), or Human polyomavirus 1, is a non-enveloped, double-stranded DNA virus that establishes lifelong latency in the urinary tract of approximately 80% of the global population. The Viral Protein 1 (VP1) is the major capsid protein, comprising 360 monomers organized into 72 pentamers. As the sole protein exposed on the virion surface, VP1 is the primary determinant of host cell receptor recognition and the exclusive target for neutralizing antibodies.

Structure and Function

The VP1 monomer features a "jelly-roll" β-barrel fold with five highly variable solvent-exposed loops: BC, DE, EF, GH, and HI. These loops facilitate the binding to α(2,3)-linked sialic acid on N-linked glycoproteins and gangliosides (e.g., GD1b, GT1b). Recent literature highlights that mutations within these loops, particularly the BC loop, allow the virus to escape host immune pressure, leading to the emergence of distinct serotypes.

Clinical Impact and Signaling

In immunocompromised individuals, particularly kidney transplant recipients, BKV reactivates and undergoes lytic replication. This process triggers inflammatory signaling pathways and can lead to:

  • Polyomavirus-Associated Nephropathy (BKVAN): Affects up to 10% of kidney transplants and often results in graft loss.
  • Hemorrhagic Cystitis (HC): A severe complication in hematopoietic stem cell transplant (HSCT) patients characterized by painful hematuria.
  • Urothelial Cancer: Emerging evidence suggests chromosomal integration of BKV in a small percentage of bladder and ureteric malignancies.

Applications of anti BK virus VP1 Neutralizing Antibodies

Neutralizing antibodies (NAbs) against VP1 are indispensable tools in both clinical diagnostics and therapeutic development, offering multifaceted utility across the biopharmaceutical pipeline:

  • Viral Entry Inhibition and Mechanism Studies: NAbs bind to specific epitopes on the VP1 loops, sterically hindering the critical interaction between the virus and host ganglioside receptors. This effectively blocks the attachment and subsequent internalization of the virion, providing a baseline for studying viral entry kinetics and cellular tropism.
  • Passive Immunotherapy Research: The passive transfer of high-titer anti-VP1 antibodies is currently a primary area of investigation for "pre-emptive" therapeutic strategies. By neutralizing circulating virions, these antibodies aim to lower viral loads in viremic patients before the onset of irreversible tissue damage or graft rejection.
  • Vaccine Development and Quality Control: VP1-based Virus-Like Particles (VLPs) are considered the most promising immunogens for BKV prevention. High-affinity antibodies serve as the essential benchmark to validate that vaccine-induced immune responses are functionally neutralizing, ensuring that the structural integrity of the VLP correlates with biological protection.
  • Serotyping & Epidemiological Assays: With four distinct BKV serotypes (I-IV) and numerous subtypes, specific antibodies are required to differentiate infections. This is vital for studying genotype-mismatch infections in transplant settings, where a donor's BKV strain may differ from the recipient's existing immunity.

Our anti BK virus VP1 Neutralizing Antibodies

Creative Biolabs' Neutra™ series represents the pinnacle of antibody engineering for polyomavirus research. Our catalog includes:

  • Pan-Serotype Neutralizers: Monoclonal antibodies (mAbs) targeting conserved epitopes at the five-fold symmetry pore or the base of the surface loops, ensuring protection against diverse BKV strains.
  • Genotype-Specific Antibodies: High-fidelity clones that can distinguish between BKV Genotype I (most prevalent) and Genotype IV (common in specific geographic regions).
  • Reporter-Labeled Variants: Biotin, FITC, or HRP-conjugated anti-VP1 antibodies for rapid implementation in existing laboratory workflows.

All Creative Biolabs products undergo rigorous validation via pseudovirus neutralization assays (PsVNA) and cell-based infection models to ensure they meet the highest standards of potency and reproducibility.
