Product list

Are you currently facing challenges in antibody development for gastrointestinal pathogens or experiencing difficulty in neutralizing the potent effects of Clostridioides difficile toxins in complex clinical models? Our C. difficile toxA Specific Neutra™ Antibody Products help you develop highly specific therapies and diagnostic tools through advanced recombinant DNA technology and high-affinity neutralizing platforms. By targeting the critical functional domains of TcdA, Creative Biolabs provides the precision required to mitigate epithelial damage and streamline your biopharmaceutical development.

Introduction of C. difficile toxA

Clostridioides difficile toxin A (TcdA) is a massive, 308 kDa exotoxin that serves as a primary virulence factor in C. difficile infection (CDI). Together with Toxin B (TcdB), TcdA is responsible for the clinical manifestations of antibiotic-associated diarrhea and pseudomembranous colitis.

Structure and Mechanism

TcdA is a multi-domain protein consisting of:

  • Glucosyltransferase Domain (GTD): The N-terminal enzymatic region that inactivates Rho family GTPases.
  • Cysteine Protease Domain (CPD): Responsible for the autocatalytic cleavage and release of the GTD into the host cytosol.
  • Transmembrane Domain (TMD): Facilitates the translocation of the toxin across the endosomal membrane.
  • Combined Repetitive Oligopeptide (CROP) Domain: The C-terminal region responsible for binding to cell-surface receptors such as glycoprotein 96 (gp96) and sulfated glycosaminoglycans.

Pathogenic Signaling

Once internalized, TcdA covalently glucosylates Rho, Rac, and Cdc42 GTPases. This modification disrupts the actin cytoskeleton, leading to "cell rounding," loss of tight junction integrity, and massive fluid secretion. The resulting inflammation and neutrophil recruitment characterize the severe mucosal damage seen in CDI.

Clinical Impact

CDI is a significant global health burden, often recurring in up to 25% of patients after standard antibiotic treatment. Because TcdA targets the intestinal epithelium directly, neutralizing its activity is a critical strategy for preventing primary and recurrent disease.

Applications of anti C. difficile toxA Neutralizing Antibodies

Neutralizing antibodies against TcdA are essential across various sectors of biopharmaceutical research:

  • Therapeutic Development: Used in combination with TcdB-targeting antibodies to provide comprehensive protection against hypervirulent C. difficile strains.
  • Passive Immunization Studies: Investigating the efficacy of oral or systemic delivery to prevent CDI recurrence in high-risk populations.
  • Diagnostic Assay Development: Serving as the capture or detection element in high-sensitivity Immunoassays to distinguish between colonization and active infection.
  • Mechanistic Research: Employed as tools to block specific toxin domains in vitro to study the kinetics of pore formation and cellular uptake.
