CD36 (CD36 Molecule), also known as platelet glycoprotein 4 or SCARB3, is a ubiquitous integral membrane protein found on the surface of multiple cell types, such as microvascular endothelial cells, phagocytes, erythroid precursor cells, skeletal muscle cells, and mammary epithelial cells. CD36 is capable of binding a variety of ligands, including collagen, thrombospondin, long-chain fatty acids as well as native lipoproteins.

CD36 has multiple biological functions. It is reported to be involved in the management of diverse metabolic pathways, such as fat taste perception, chylomicron production, and eicosanoid formation. In addition, CD36 is also considered to be a hub for parenchymal cell fatty acid uptake and regulates glucose and insulin metabolism. Another crucial function of CD36 is to directly mediate cell adhesion of erythrocytes. In addition, as a regulator of angiogenesis, CD36 is also closely linked to tumor growth and wound healing.
Increasing evidence suggests that the function of CD36 is associated with the activation of src family kinases as well as the mitogen-activated protein (MAP) kinases. In addition, there are data suggesting that CD36-mediated platelet activation is linked to the MAP kinase pathway.

CD36 is a critical regulator of fatty acid metabolism, heart disease, and intestinal fat processing. Increasing evidence suggests that CD36 may be closely linked to a variety of diseases, including atherosclerosis, glucose intolerance, diabetes, arterial hypertension, cardiomyopathy, and Alzheimer's disease. In addition, CD36 also serves as an important factor in the occurrence of various cancers, such as liver cancer and glioma. It is noteworthy that CD36 also participates in the regulation of inflammatory responses and atherothrombotic disease. It has been reported that CD36 may also be a promoter of obesity.
Function-blocking antibodies targeting CD36 have been reported to effectively reduce tumor metastasis in gastric and ovarian cancer mice. Furthermore, targeting CD36 has been shown to elicit antitumor immunity in immunocompetent mice, thus providing new horizons for CD36-targeted therapy in cancer.
