CD40LG Specific Neutra™ Antibody Products
Product listCD40 ligand (CD40LG) is a cytokine, also known as CD40L. The membrane-bound CD40LG is a single-pass type II membrane protein mainly expressed on activated CD4+ T cells and induces apoptosis. Upon binding with the CD40 receptor, the membrane-bound CD40LG is cleaved into a soluble form. The soluble form protein is secreted intracellularly and plays a role in cell death induction. CD40LG interacts with CD40, a member of the TNF-receptor superfamily. The CD40-CD40LG signaling cascade transduces various cell type-specific immune responses.

Signal Pathways and Receptors for CD40LG
As shown in Figure 1, CD40LG interacts with its receptors such as αMβ2, α2bβ3, α5β1, and its "classical" receptor CD40. The trimeric D40LG complexes interact with CD40 in a homotrimer or heterotrimer fashion. CD40LG induces cell type-specific downstream signaling events including the recruitment of TRAFs and the engagement of NF-κB, MAPKs, and PI3K.

Clinical Significance of CD40LG
Due to its indispensable role in B cell activation and antibody production, CD40LG is a compelling signal pathway modulator in inflammation and autoimmune diseases. Treatment strategies based on CD40LG are currently being investigated for several diseases:
- sCD40LG is a biomarker for diseases such as myocardial infarction, virus inflammation, and Alzheimer's disease.
- CAR-T cells incorporated with CD40LG may elicit enhanced endogenous antitumor responses in cellular immunotherapy.
- Anti-CD40LG antibodies may specifically block the activation of CD40LG-associated immune function in cardiovascular disease.
