Tissue Factor or Coagulation Factor III (F3) is a glycoprotein situated on the cellular surface. F3 facilitates the initiation of blood coagulation cascades and serves as the high-affinity receptor for coagulation factor VII. F3 comprises three distinct domains: cytoplasmic, extracellular, and transmembrane domain. Notably, F3 is unique in the coagulation pathway as no congenital deficiency has been reported for it. In various cancer types, F3 contributes to progression, invasion, metastasis, and angiogenesis through activation of signaling pathways. The F3 proteolytic pathway, facilitated by factor VIIa, influences angiogenesis. Tumor cell surface F3 expression elevates VEGF expression, promoting angiogenesis. F3's cytoplasmic domain activates GTPase, Rac-1, MAPK, and p38, facilitating tumor cell migration. Additionally, F3's cytoplasmic domain regulates VEGF expression and calcium pumps in monocytes.

Numerous investigations have evaluated the anti-tumor efficacy of ADCs across diverse solid tumor types.
AsTF, a secreted variant of F3, triggers blood coagulation in patients exhibiting elevated F3 expression levels in various solid tumors like pancreatic ductal adenocarcinoma (PDAC). Consequently, a neutralizing antibody specific to AsTF holds the potential to impede the spread and advancement of PDAC.

Thromboembolic complications are commonly linked with advanced cancer stages. F3 is known to be upregulated in various tumor types and is present on both tumor cells and tumor vasculature. While the precise mechanisms remain unclear, F3 expression on tumor cells can initiate intracellular signaling cascades via diverse pathways, ultimately promoting tumor angiogenesis, proliferation, and metastasis. Prophylactic treatment with a humanized anti-human F3 antibody has demonstrated efficacy in inhibiting experimental lung metastasis of human breast carcinoma cells and reducing tumor incidence and growth in orthotopically implanted breast carcinoma cells.
