Integrin alpha V (ITGAV), also known as CD51 or VTNR, is an important member of the integrin alpha chain family. Integrins are closely associated with cell surface signaling. ITGAV integrins are receptors for a variety of proteins, including fibronectin, vitronectin, fibrinogen, prothrombin, and osteomodulin. They can efficiently recognize RGD sequences in a variety of ligands. ITGAV is a subunit that makes up the integrin receptor and can pair with the β1, β3, β5, β6, or β8 subunits to form the αVβ1, αVβ3, αVβ5, αVβ6, and αVβ8 subtypes. These subtypes participate in the regulation of angiogenesis through different pathways. It has been reported that ITGAV-specific antibodies can block the binding of CD51 to vitronectin and fibronectin in a variety of cell types and are effective in blocking cytotoxicity in specific cells.

ITGAV is a crucial hub for multiple cell signaling pathways, including the NRG1-ERBB signaling pathway, the FGF1 signaling pathway, and the IL1B signaling pathway. In addition to playing an integral role in cell signaling, ITGAV also serves as a necessary mediator in cell adhesion. Furthermore, ITGAV is also an important regulator of epidermal generation and maintenance. In summary, ITGAV participates in the regulation of cell growth and differentiation through interactions with various intramembrane and intracellular molecules.
ITGAV has been proven to be a crucial promoter of tumor progression in multiple tumor entities. Overexpression of ITGAV has been associated with increased metastasis and invasiveness in multiple cancers, including colorectal cancer, melanoma, prostate cancer, and breast cancer. Furthermore, ITGAV is shown to be involved in transendothelial migration of aggressive human cancers.
