KIT Proto-Oncogene, Receptor Tyrosine Kinase, also known as SCFR or CD117, is a vital member of the cytokine receptor family. The typical form of the protein has an N-terminal extracellular domain, a special C-terminal intracellular domain as well as a transmembrane domain. After activation by specific factors, this protein will phosphorylate a variety of intracellular proteins, thereby participating in cell proliferation, migration, and apoptosis. Mutations in the KIT gene may lead to the occurrence of a variety of diseases, such as mast cell disease, thymic cancer, acute myeloid leukemia, gastrointestinal stromal tumors, leiomyosarcoma, and piebaldism.

KIT has been found to be expressed on the surface of bone marrow-derived stem cells as well as on stem cells in other organs, such as the heart and liver. These shreds of evidence reveal that the KIT signaling pathway may be involved in the stemness regulation of some organs. In addition, it has been reported that KIT is a crucial regulator of various life activities, such as cell survival, mast cell development, gametogenesis, and melanin production. Furthermore, KIT signaling is involved in regulating oogenesis and spermatogenesis. Therefore, it is closely associated with female and male fertility.
KIT can activate multiple signaling pathways. KIT participates in the activation of the AKT1 signaling pathway through phosphorylation of PIK3R1. Furthermore, KIT contributes to the activation of multiple transcription factors, including STAT1, STAT3, STAT5A, and STAT5B. It is worth noting that activated KIT further performs signal transduction by regulating the activation of MAPK1/ERK2 or MAPK3/ERK1.

Overexpression or mutation of KIT protein is closely related to the occurrence of cancer.
