MET Proto-Oncogene, Receptor Tyrosine Kinase, also known as DFNB97 or RCCP2, is a vital member of the tyrosine kinase family. In addition to epithelial cells, MET is expressed in a variety of other cell types, such as hematopoietic cells, neurons, hepatocytes, and endothelial cells. Hepatocyte growth factor (HGF) is a crucial ligand for MET. Information transfer from the extracellular matrix to the cytosol is accomplished by the binding of MET to HGF. Furthermore, the HGF/MET axis is closely related to cardiac muscle development.

MET participated in many physiological processes, including survival, migration, differentiation as well as proliferation. MET is also an indispensable regulator of mitosis and morphogenesis. It is noteworthy that MET is required for critical processes such as organ regeneration and wound healing in adulthood. Ligand-bound MET provides docking sites for downstream signaling molecules. Upon activation by ligands, MET recruits downstream effectors that lead to the activation of multiple signaling cascades, including RAS-ERK and PI3 kinase-AKT. For example, GAB1 is an essential coordinator of the cellular response to MET. Upon interaction with MET, GAB1 is phosphorylated and recruits many signaling effectors, including PI3K, SHP2, and PLC-γ. Furthermore, phosphorylated GAB1 generates sustained signals that mediate most downstream signaling pathways. It has been reported that the HGF/MET axis is closely related to a variety of signaling pathways, including MAPK, PI3K, and STAT signaling pathways.

Amplification and overexpression of MET are crucial drivers of human cancer. Abnormal MET activation can trigger tumor growth and metastasis. MET-associated diseases include deafness, renal cell carcinoma, hepatocellular carcinoma, breast cancer as well as various head and neck cancers.
