Notch Receptor 3 (Notch3), also known as Neurogenic Locus Notch Homolog Protein 3 or CADASIL, is an essential member of the Notch family. The Notch3 protein serves as a trigger across the cell membrane. One end of the Notch3 remains inside the cell and the other end protrudes from the surface of the cell. Some ligands bind to the outside of Notch3, which causes the ends of the Notch3 protein to dissociate within the cell. The separated fraction enters the nucleus to regulate gene expression.

Notch3 serves as a signaling receptor involved in the regulation of diverse developmental and cellular life events. Notch3 signaling is closely linked to stem cell survival and proliferation during embryonic development. In adults, Notch3 not only plays a crucial role in stem cell regulation but is also involved in regulating the survival and function of vascular smooth muscle cells. Notch3 also participates in the regulatory process of esophageal homeostasis. Furthermore, Notch3 has been reported to be a critical regulator of proinflammatory macrophage development.
The Notch3 signaling pathway is evolutionarily highly conserved. The Notch3 signaling pathway plays a crucial role in the development and cell specification of many organs. It has been reported that the Notch3 signaling pathway is one of the most commonly activated signaling pathways in cancer. Notch3 has been reported to play an essential function in activating NF-κB.
