Western blot analysis of V3S-0622-YC5705 (Fc silenced) was performed by loading human CLEC2D protein (CLD-H5253, acrobiosystems) onto a 12% Tris-HCl polyacrylamide gel. Proteins were transferred to a CN membrane and blocked with 5% skim milk for at least one hour. Membranes were probed with V3S-0622-YC5705-Fc silenced and HRP goat anti-Human IgG as a secondary antibody (1: 2000). Chemiluminescent detection was performed.
Lane 1: Reducing antigen (0.1 μg)

ELISA analysis of V3S-0622-YC5705 (Fc silenced) was performed by coating with human CLEC2D protein (CLD-H5253, acrobiosystems). Then blocked with BSA and incubated with V3S-0622-YC5705-Fc silenced. The HRP-conjugated goat anti-Human IgG as a secondary antibody (1: 2000). Detection was performed using TMB substrate and stopped with sulfuric acid. The absorbances were read on a spectrophotometer at 450 nm.

Flow cytometry analysis of Daudi cells with purified antibody V3S-0622-YC5705-Fc silenced (TE221587-2, Blue). Cells were incubated with V3S-0622-YC5705-Fc silenced (5 μg/5x10^5 cells) for 30 min at 4°C. The secondary antibody used was FITC-Goat Anti-Human IgG Fc antibody (ab97224, abcam) for 30 min at 4°C.
