Application Notes | S trimer and RBD ELISAs were performed in triplicate. 175 ng of antigen per well was coated onto 96-well ELISA plates at 4°C overnight. Plates were washed and then blocked with 100 μL of blocking buffer at 37°C for 2 hr. Purified antibodies were serial diluted using dilution buffer, added to the antigen-coated blocked plates, and then incubated at 4°C for 1 hr. Plates were washed and 50 μL of a secondary anti-human kappa light chain detection antibody was added to each well and incubated at room temperature for 1 hr. After the final wash, 50 μL TMB substrate was used to detect antibody binding to antigen measuring absorbance at 405 nm. The antibody 910-30 showed an IC50 of 0.071 μg/mL against a vesicular stomatitis virus SARS-CoV-2 pseudovirus and 0.142 μg/mL against authentic SARS-CoV-2. |
