Application Notes | Ninety-six half area well-plates were coated overnight at 4°C with 25 μl of sarbecovirus RBD proteins, prepared at 5 μg/ml in PBS pH 7.2. Plates were then blocked with PBS 1% BSA and subsequently incubated with mAb serial dilutions for 1 h at room temperature. After 4 washing steps with PBS-0.05% Tween 20, goat anti-human IgG secondary antibody was added and incubated for 1 h at room temperature. Plates were then washed four times with PBS-T and 4-nitrophenyl phosphate substrate was added. After 30 min incubation, absorbance at 405 nm was measured by a plate reader and data were plotted using Prism GraphPad. |
