Coagulation factor XI (F11) is an important component of the blood coagulation cascade. It is also known as FXI. As a secretory protein, F11 is overexpressed in normal tissues such as the liver, serum, plasma, and pancreas. F11 is associated with Factor Xi Deficiency and Thrombocytopenia diseases.

Effects of Targeting F11 in Thrombosis

Treatment Strategies Associated with F11

F11 inhibitors could safely decrease the thrombus size, produce antithrombotic activity, and reduce ischemic events. Therefore, compounds targeting F11 have become promising approaches to prevent and treat thrombosis.

Antisense oligonucleotides (ASOs) synthesize proteins intracellularly to block F11 with longer half-life times.
Anti-F11 antagonist monoclonal antibodies have been used in studies for the prevention of thrombosis.
Small peptidomimetic molecules, delivered orally, have a short half-life in company with renal clearance.
Natural inhibitors may have a slow on/offset and a relatively high risk.
