Product listFibroblast growth factor receptor 2 (FGFR2), also known as CD332 or KGFR, is a transmembrane catalytic receptor with intracellular tyrosine kinase activity. Due to alternative splicing, FGFR2 has multiple isoforms, of which FGFR2b and FGFR2c are two representative ones.

FGFR2 Signaling Pathway
As a receptor for fibroblast growth factors, the interaction between FGFR2 and fibroblast growth factors induces a cascade of downstream signals. FGFR2 transduces FGF signals to signaling cascades such as ERK, PI3K-AKT, and IP3-DAG.
- The FGF-PI3K-AKT signaling pathway is involved in cell survival and polarity regulation.
- The FGF-RAS-ERK signaling pathway is involved in cell proliferation.
- The FGF-IP3-DAG signaling pathway is involved in cellular growth and metabolism.

Abnormalities of FGFR2
In the absence of fibroblast growth factors, the wild type of FGFR2 is autoinhibited by the kinase hinge. Missense mutations of FGFR2 (see Fig.2) may result in altered ligand affinity, leading to congenital diseases such as skeletal disorders and cancers.
FGFR2 fusion proteins, due to the chromosomal translocation, have been identified as another important carcinogenic factor in various cancers such as cholangiocarcinoma.

Clinical Significance of FGFR2
Tab. 1 provides a list of inhibitors targeting FGFR2 fusion proteins or mutations that are in preclinical and clinical trials. Tyrosine kinase inhibitors (TKI) targeting FGFR2 alone or in combination therapies are also listed here.
