MSLN is overexpressed in several types of solid tumors and participates in cell adhesion, tumor progression, metastasis, and drug resistance. So it is considered a tumor marker and has received considerable attention in the field of cancer research.

Role of MSLN in Cancers and Signaling
MSLN is highly expressed in several types of solid tumors, particularly mesothelioma, pancreatic cancer, ovarian cancer, and lung adenocarcinoma. It plays a potential role in cell adhesion, tumor progression, metastasis, and drug resistance. The interaction between MSLN and its widely recognized binding partner, CA125/MUC16, may lead to tumor implantation and peritoneal spread via cell adhesion. Furthermore, MSLN has been shown to activate several signaling pathways, including the NF-κB pathway, MAPK/ERK signaling pathways, and the PI3K/AKT pathway, which overall result in the inhibition of apoptosis and increased cell survival.

Anti-MSLN Antibody-Based Therapy
Over the last two decades, many types of novel anti-MSLN antibody-based therapies have been developed such as monoclonal antibodies, ADCs, and immunotoxins. The action of antibody-based drugs involves several mechanisms, including antibody-mediated neutralization of the tumor-associated antigen (TAA) resulting in complement-mediated killing of the tumor cell; antibody-dependent cellular cytotoxicity (ADCC) via NK cells resulting in tumor cell death; antibody-dependent cellular phagocytosis (ADCP) of the tumor cells via monocyte-derived macrophages, and lastly inhibition of tumor cell proliferation and induction of apoptosis via TAA targeting utilizing antibodies linked to poisons or inhibitors. Multiple drugs for targeted therapy have been created based on the murine-derived MSLN antibody or other modified and humanized types. Clinical trials have shown that anti-MSLN antibodies can improve outcomes in patients with mesothelioma and pancreatic cancer.

Neutralizing Antibodies Targeting MSLN
The neutralizing antibodies are developed for both research and diagnostic purposes. These antibodies have been extensively validated and characterized to ensure high specificity and sensitivity in detecting MSLN in different biological samples, such as tissue sections, cell lines, and body fluids. They can be used to visualize and quantify MSLN expression in tumor tissues, allowing us to study its role in cancer progression and prognosis.
