Carbonic anhydrase 9 (CA9), also known as CAIX or MN, is a common transmembrane glycoprotein. CA9 is a critical member of the tumor-associated carbonic anhydrase isoenzyme. In addition, CA9 is highly expressed in the gastrointestinal tract and can promote gastric acid secretion. As a hypoxia-inducible protein, CA9 is present in many human cancers but absent in most normal tissues.

Functions

CA9 has a high catalytic activity and therefore plays a crucial role in tumor acidification. CA9 is also an essential factor in catalyzing the reversible hydration of carbon dioxide. In addition, CA9 may be closely related to a variety of biological processes, including calcification, bone resorption, acid-base balance, respiration, and the production of aqueous humor, saliva, and gastric acid. Most importantly, CA9 may also mediate cell proliferation and transformation.

CA9 and Carcinoma

CA9 is proven to be abnormally expressed in a variety of cancers, including renal cell carcinoma, esophageal cancer, cervical cancer, lung cancer, colon cancer, breast cancer, and brain cancer. The overexpression of CA9 in cancer tissues is closely linked to the hypoxic conditions in the tumor microenvironment caused by abnormal vascular systems. Due to the differential expression of CA9 in cancer and normal tissues, CA9 has gradually become a powerful target for the diagnosis of various tumors.

Therapies Targeting CA9

Currently, CA9 is regarded as a promising target for imaging and treatment of various hypoxic tumors. Therefore, a variety of highly effective and selective CA9 inhibitors have been gradually developed, including antibodies and small molecules.

CA9 Signaling Pathways

  • NF-κB pathway

It has been reported that CA9 is essential for the activation of NF-κB under hypoxic conditions. The interaction between CA9 and NF-κB may affect cell proliferation, apoptosis, and metastasis.

  • MEK/Raf/ERK pathway

Abnormal expression of CA9 leads to phosphorylation of extracellular signal-regulated kinase (ERK), thereby inducing cell migration. In addition, increasing evidence suggests that the MEK/Raf/ERK signaling pathway is required for CA9-induced epithelial-mesenchymal transition.
