The CD8a molecule, also known as CD8 alpha chain, is a glycoprotein expressed on the surface of cytotoxic T cells, natural killer cells, and some dendritic cells. These molecules aid T cell recognition of antigens via major histocompatibility complex class I molecules, strengthening T cell activation and cytotoxicity. CD8a-related signaling pathways activate protein kinases, triggering transcription factors like NF-κB and AP-1. CD8a is critical in promoting T cell responses against abnormal cells, pathogen clearance and immunity defense enhancement. Diseases associated with CD8a include CD8 deficiency, familial and primary cutaneous aggressive epidermotropic CD8+ t-cell lymphoma. Understanding the function and regulation of CD8a is essential for elucidating immune responses and developing immunotherapeutic strategies for various diseases.

Physiological Functions of CD8a

CD8a, a key component of the CD8 T-cell co-receptor complex, plays essential roles in immune surveillance, defense mechanisms, and maintaining immune homeostasis. Its physiological functions are diverse and crucial for orchestrating effective immune responses.

Firstly, CD8a is primarily expressed on cytotoxic T cells (CTLs), where it acts as a co-receptor for the T cell receptor (TCR). Upon recognition of antigenic peptides presented by major histocompatibility complex class I (MHC-I) molecules on target cells, CD8a enhances the binding affinity of the TCR to the peptide-MHC-I complex, promoting T cell activation and cytotoxicity. This mechanism enables CTLs to identify and eliminate infected or aberrant cells, thereby contributing to immune surveillance and host defense against pathogens and cancer.

Furthermore, CD8a is involved in T cell differentiation and effector functions. It plays a role in the development of memory T cells, which provide long-lasting immunity upon re-exposure to specific antigens. CD8a-expressing memory T cells exhibit enhanced responsiveness and rapid effector functions, contributing to the rapid clearance of pathogens during secondary immune responses.

Moreover, CD8a is implicated in immune regulation and tolerance. It is involved in the formation of immunological synapses between T cells and antigen-presenting cells (APCs), where it modulates T cell activation thresholds and signaling cascades. Additionally, CD8a-expressing regulatory T cells (Tregs) play a role in immune tolerance and preventing autoimmunity by suppressing excessive immune responses.

  • Immuno-PET monitoring of lymphocytes using the CD8a-specific antibody

The radioactive labeling of CD8a-specific antibodies enables non-invasive imaging and tracking of CD8+ T cells, thereby achieving immune-PET monitoring of lymphocytes in cancer treatment. This approach aims to overcome limitations of traditional biopsy and provide a comprehensive view of the immune cell dynamics within the tumor microenvironment across time and different tissue sites. It holds the potential to identify responders and non-responders early in the treatment process, adjust therapeutic strategies promptly, and better understand the dynamics of immune infiltration and tumor response. CD8a antibody plays a pivotal role in the development of novel diagnostic tools like immuno-PET for cancer immunotherapy.

  • CD8a as a Prognostic and Immunotherapy Predictive Biomarker
    High expression levels of CD8a correlate with better survival outcomes, suggesting its role as a protective gene. CD8a is also a predictive biomarker for immunotherapy response. The study shows that low expression of CD8a is related to poor outcomes in bladder cancer patients treated with immunotherapies, such as PD-1 inhibitors. This indicates that CD8a could potentially be used to identify patients who are more likely to benefit from such treatments. The expression of CD8a has a high correlation with the presence of certain types of tumor-infiltrating immune cells (TIICs), such as CD8+ T cells and M1 macrophages. This correlation underscores the role of CD8a in influencing the immune landscape within the tumor microenvironment, which is crucial for the effectiveness of immunotherapy.
