High-temperature requirement A serine peptidase 1 (HTRA1) belongs to the HtrA family of serine proteases and plays a part in several biological processes. HTRA1 is typically secreted into the extracellular space and can also be found in various tissues and body fluids. Structurally, HTRA1 consists of a trimeric arrangement of protease domains with a PDZ domain at the C-terminus. Functionally, HTRA1 plays a role in the regulation of extracellular matrix components, cell signaling pathways, and apoptosis. Additionally, HTRA1 has been recognized for its role in tissue remodeling and its potential as a biomarker and therapeutic target in various pathological conditions.

HTRA1 participates in several signaling pathways that regulate biological activities. Additionally, HTRA1 has been linked to the regulation of Wnt signaling pathways, which are critical for cell proliferation and differentiation. Moreover, HTRA1 can interact with components of the Notch signaling pathway, influencing cell fate determination and tissue homeostasis. In cancers, HTRA1's dysregulation has been observed in several malignancies, including breast, lung, ovarian, and pancreatic cancers. It has been linked to tumor progression, invasion, metastasis, and drug resistance, making it a potential prognostic biomarker and therapeutic target.
In cerebrovascular diseases, particularly in cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL), HTRA1 has been associated with vascular dysfunction and the pathogenesis of the disease.
Moreover, HTRA1's involvement extends to other conditions such as age-related macular degeneration (AMD), where genetic variations in the HTRA1 gene have been linked to an increased risk of developing AMD. In osteoarthritis, HTRA1 has been implicated in cartilage degradation and joint inflammation, suggesting a role in the disease's progression. Additionally, HTRA1 dysregulation has been reported in neurodegenerative disorders like Parkinson's disease and Alzheimer's disease, indicating its potential involvement in protein misfolding and aggregation processes.
