Product listProgrammed cell death protein 1 (PDCD1), also named as PD-1 or CD279, is a protein expressed in immune cells such as T cells, B cells and NK cells. PD-1 is an immune checkpoint protein involved in apoptosis of T cells. As a drug target, the inhibitors against PD-1 can attack tumors by blocking PD-1 interaction via activating the immune system.
Function
As an inhibitor, PD-1 plays an important role in the immune system, including down-regulating the immune responses and promoting self-tolerance. Here are some functions that PD-1 may involve in.
- Regulate T-cell functions.
- Promotes the function and development of Tregs.
- Inhibits the function of cytotoxic T lymphocytes.
- Protect against autoimmunity.
- Reduce antimicrobial immunity.
- Attenuate antitumor immunity.
Signaling pathways targeted by PD-1
The ligands of PD-1 are mainly known as PD-L1 and PD-L2. PD-1 with its ligands are involved in a number of signaling pathways to exert immunosuppressive functions in T cells.
- PD-1:PD-L1 pathway
- PI3K/AKT pathway
- Mitogen-activated protein kinase (MAPK) pathway
- Mammalian target of rapamycin (mTOR) pathway
Clinical significance of PD-1
PD-L1 is highly expressed in several cancers and blockade of PD-1/PD-L1 interaction can enhance T-cell responses and mediate preclinical antitumor activity. Therefore, PD-1 is a promising target for immunotherapies and anti-PD-1 blocking antibodies are being developed for immunotherapies of cancer and chronic infections.
