EGFR Signaling Pathways
Activation by ligands prompts EGFR to initiate a time-dependent sequence of molecular events, including the downregulation of numerous microRNAs, upregulation of newly synthesized mRNAs, and covalent modifications of proteins. These processes collectively control the expression of genes that determine cellular phenotypes. Circular RNAs and long non-coding RNAs are involved in EGFR signaling, complementing the regulatory functions of microRNAs. EGFR, in conjunction with driver mutations, actively contributes to metastasis through diverse mechanisms. Paracrine loops involving interactions between stromal and tumor cells provide the necessary support for EGFR in promoting the survival of circulating tumor cell clusters, facilitating the colonization of distant organs, and enabling invasion across tissue barriers.
