Product listSynuclein alpha (SNCA) is also known as α-Synuclein and α-Syn. SNCA is a neuronal protein and has three isoforms because of alternatively splice. The soluble SNCA is secreted to integrate presynaptic signaling and the membrane-bound SNCA may serve for membrane trafficking in the cytoplasm, nucleus, and synapse. Dysfunction and aggregation of SNCA are associated with neurological disorders such as Parkinson's disease (PD) and dementia with lewy bodies (DLB).

Pathological Mechanisms of SNCA Aggregation
The monomers, oligomers, and fibrils are different forms of SNCA. In healthy situations, SNCA is almost soluble and in a smaller size. But in pathogenic conditions, SNCA tends to function in the aggregated forms such as oligomers and fibrils. There are several pathological mechanisms associated with SNCA oligomerization, fibrillization, and aggregation.
- Promotes the amyloid formation which may compromise the homeostasis of neuronal cells.
- Triggers the apoptosis and dysfunction of neuronal cells.
- Elicits selective neurodegeneration and neurotoxicity and results in neuroinflammation.
- Induces innate and adaptive immunity including the activation of immune cells.

Targeting SNCA Strategies for PD
Treatment strategies of PD have been developed based on the SNCA aggregation kinetics and its genetic factors.
- Active immunization with SNCA antigens in the form of vaccines to help produce SNCA antibodies.
- Passive immunization with antagonist antibodies against extracellular SNCA preventing its spread.
- Reduction of the expression of SNCA with small molecules that reduce SNCA levels by inhibiting the mRNA translation.
- Inhibition of the aggregation of SNCA with dis-aggregators to block the self-assembly and amyloid formation.
- Targeting SNCA pro-aggregation factors.
- Decreasing synaptic activity.
